Description 

The 13810-14507 aa of human MUC16 coding sequences was inserted to mouse Muc16 gene locus in B-hMUC16 ID8 cells, truncated human MUC16 was highly expressed on the surface of B-hMUC16 ID8 cells. It is recommended to do binding experiments before use.

Application

B-hMUC16 ID8 cells have the capability to establish tumors in C57BL/6 mice in vivo and can be used for efficacy studies.



Targeting strategy



Gene targeting strategy for B-hMUC16 ID8 cells. The exogenous promoter and 13810-14507aa human MUC16 CDS was inserted to mouse Muc16 gene in B-hMUC16 ID8 cells.

MUC16 expression analysis in B-hMUC16 ID8 cells by flow cytometry. Single cell suspensions from B-hMUC16 ID8 cultures were stained with species-specific anti-MUC16 antibody. Human MUC16 were detected on the surface of B-hMUC16 ID8 cells. The 1-F11 clone of B-hMUC16 ID8 cells was used for in vivo experiments.

Subcutaneous homograft tumor growth of B-hMUC16 ID8 cells. B-hMUC16 ID8 cells (5x105) and wild-type ID8 cells (5x105) were subcutaneously implanted into C57BL/6 mice (female, 8-week-old, n=6). Tumor volume and body weight were measured twice a week. (A)  Body weight (Mean± SEM). (B) Average tumor volume ± SEM. Volume was expressed in mm3 using the formula: V=0.5 X long diameter X short diameter2. As shown in panel A, B-hMUC16 ID8 cells were able to establish tumors in vivo and can be used for efficacy studies.
